Dimitris Zografakis put the home side ahead in the 11th minute when he met a corner from Brazilian Luciano.
Former Olympiakos player Luciano, who was making his debut for Panionios after joining from Xanthi, doubled the lead with a powerful shot in the 19th minute.
Olympiakos struck back six minutes later when Konstantinou flicked in a header by Argentine defender Gabriel Schurrer.
Olympiakos captain Predrag Djordjevic pulled his side level, stroking home a 39th minute penalty after Dimitris Mavrogenidis was brought down.
